I have little knowledge of how the insides of the gameboy work, but I was wondering if instead of just wiring an LED behind the speaker grill in a DMG, it would be possible to take ou the speaker and wire an LED onto the speaker's wires. I get the feeling this wouldn't work either the way I imagine or at all, but has anyone tried?

Are you trying to achieve sound-reactive LEDs? That's what you'll get if you attach an LED to the wires, but the LED will likely be very dim and may not even light up at all.

That is exactly what I'm looking for. Would I need to put a resistor on it? From the sound of it, the thing won't be getting more power than it can handle.

Relying on the amplifier for the headphone jack? Not a bad idea... not great, but not bad.
(it still won't be very bright)

You'll probably need some sort of op-amp to boost the signal if you want to drive an LED.

i actually did this for a project at school. but i used an ic and like 8 or 9 leds and transistors =P look into the lm389 IC
